چکیده

Breast cancer is one of most common malignant tumor in global female. And the leading cause of morbidity and mortality in breast cancer patients is cancer metastases, but not the primary tumor (Porter, 2009; Sanchez-Zamorano et al., 2011). It has been demonstrated that tumor cell invasion is the key event in the metastatic steps (Geiger et al., 2009). However, recent studies show that cancer progression toward metastasis is not entirely dependent on tumor cells themselves, but the result of the interactions of tumor cells and the primary tumor microenvironment (Allen et al., 2011). Cancer cells, specially those at the edges of the tumor, can attract and stimulate host cells to secret many kinds of factors (TGF-β1, IL-1, TNF-α, etc.) in the tumor microenvironment. Those factors can generate a series of invasion-related biological effects through the tumor cell signal transduction pathways, and then cause the cells to separate from the primary lesion focus and result in local invasion and distant mestastasis finally (Allen et al., 2011). So far, novel findings indicates that TGF-β1 is one of the factors which could be highly

Wogonoside inhibits invasion and migration through suppressing TRAF2/4 expression in breast cancer

BACKGROUND Twist1 is involved in tumor initiation and progression, which especially contributes to tumor invasion and metastasis. Wogonoside is the main in-vivo metabolite of wogonin, and it is also a natural product with potential treatment effects against cancer. METHODS In this study, we investigated the in-vitro anti-invasion and in-vivo anti-metastasis effects of wogonoside on breast can...

Curcumin Inhibits Transforming Growth Factor-β1-Induced EMT via PPARγ Pathway, Not Smad Pathway in Renal Tubular Epithelial Cells

Tubulointerstitial fibrosis (TIF) is the final common pathway in the end-stage renal disease. Epithelial-to-mesenchymal transition (EMT) is considered a major contributor to the TIF by increasing the number of myofibroblasts. Curcumin, a polyphenolic compound derived from rhizomes of Curcuma, has been shown to possess potent anti-fibrotic properties but the mechanism remains elusive. We found t...

Sarsaparilla (Smilax Glabra Rhizome) Extract Inhibits Migration and Invasion of Cancer Cells by Suppressing TGF-β1 Pathway

Sarsaparilla, also known as Smilax Glabra Rhizome (SGR), was shown to modulate immunity, protect against liver injury, lower blood glucose and suppress cancer. However, its effects on cancer cell adhesion, migration and invasion were unclear. In the present study, we found that the supernatant of water-soluble extract from SGR (SW) could promote adhesion, inhibit migration and invasion of HepG2...

CK2 Inhibitor CX-4945 Blocks TGF-β1-Induced Epithelial-to-Mesenchymal Transition in A549 Human Lung Adenocarcinoma Cells

BACKGROUND The epithelial-to-mesenchymal transition (EMT) is a major phenotype of cancer metastasis and invasion. As a druggable cancer target, the inhibition of protein kinase CK2 (formally named to casein kinase 2) has been suggested as a promising therapeutic strategy to treat EMT-controlled cancer metastasis. This study aimed to evaluate the effect of the CK2 inhibitor CX-4945 on the proces...

Berberine suppresses TNF-alpha-induced MMP-9 and cell invasion through inhibition of AP-1 activity in MDA-MB-231 human breast cancer cells.

Invasion of cancer cell induced by matrix metalloproteinase-9 (MMP-9) is one of pivotal steps in cancer metastasis. Herein, we investigated how cell invasion was regulated by berberine (BBR), an isoquinoline derivative alkaloid compound, in MDA-MB-231 human breast cancer cells. The basal level of MMP-9 activity and expression was dose-dependently increased by TNF-alpha, while TNF-a-induced MMP-...
